         Can Molecular Simulations help us understand how communicate?

                                   Abstract:
 Neurotransmitter-gated ion channels are complex neuroreceptors located in the
 membrane of nerve cells that control the rapid transmission of nerve impulses.
   They are involved in many neurological disorders, such as Alzheimer`s and
 Parkinson`s diseases, and are the sites of action of drugs such as valium and
   many anaesthetics. However we have little idea of how they function at the
   molecular level. In particular it is not clear how the binding of a small
molecule (the neurotransmitter) triggers a series of events culminating into the
 opening (gating) of the transmembrane channel: ions can then flood across the
    cell membrane modifying the cell activity. We use a range of simulations
 techniques, including classical molecular dynamics, density functional theory
     and the metadynamics method, in combination with data from mutagenesis
       experiments to elucidate at the atomistic level the mechanisms of
neurotransmitter binding and of channel gating in two neurotransmitter-gated ion
                 channels, the 5-HT_3 and the GABA_C receptors.
